EazeQuant Mouse TNF alpha ELISA Kit is used to quantitatively detect mouse TNF alpha (TNF-α) in cell culture supernatant, serum and EDTA/citrate plasma.

In the TNF superfamily, TNF-α is a pro-inflammatory cytokine mainly secreted by macrophages, binding to its receptors TNFRSF1A/TNFR1 and TNFRSF1B/TNFBR. It plays a role in regulating immune cells, cell proliferation, differentiation, apoptosis, lipid metabolism and coagulation. Showing as a 17 kDa band on SDS-PAGE in non-reducing conditions, TNF-α exists as a multimer of two, three or five non-covalently linked units. The studies of TNF-α in mice suggest its functions in neuroprotection and tumor necrosis. TNF-α is also involved in the immune responses against various pathogens and the necrosis of specific tumors. Being synergistic with IFN-γ, it induces cytolysis or cytostasis of certain transformed cells. TNF-α is associated with autoimmune diseases, insulin resistance and cancer.

In the assay, samples, standards or controls are added to the microplate coated with the primary antibody, and then bind to the immobilized antibody in the microplate wells. Subsequently, the second antibody is introduced to the mixtures as the detector, followed by the addition of a substrate solution that generates signal to be measured. The intensity of this signal correlates to the concentration of mouse TNF-α in the original specimen.
