EazeQuant Mouse IL-10 ELISA Kit is used to quantitatively detect mouse Interleukin-10 (IL-10) in cell culture supernatant, serum and plasma.

IL-10 is a cytokine protein mainly produced by monocytes, with some production by lymphocytes. It plays a critical role in immunoregulation and inflammation. IL-10 exerts its effects by down-regulating the expression of Th1 cytokines, MHC Class II antigens, and costimulatory molecules on macrophages. It enhances B cell survival, proliferation, and antibody production. In addition, IL-10 inhibits the activity of NF-κB and is involved in regulating the JAK-STAT signaling pathway. Studies in mice have highlighted the essential role of IL-10 as an immunoregulator in the intestinal tract. The mutations in the IL10 gene have been associated with an increased susceptibility to HIV-1 infection and rheumatoid arthritis. Diseases linked to IL10 include Graft-Versus-Host Disease and HIV-1.

In the assay, samples, standards or controls are added to the microplate coated with the primary antibody, and then bind to the immobilized antibody in the microplate wells. Subsequently, the second antibody is introduced to the mixtures as the detector, followed by the addition of a substrate solution that generates signal to be measured. The intensity of this signal correlates to the concentration of mouse IL-10 in the original specimen.
