EazeQuant Human TNF alpha ELISA Kit is used to quantitatively detect human Tumor Necrosis Factor α (TNF-α) in cell culture supernatant, serum, plasma and urine.

In the TNF superfamily, TNF-α is a pro-inflammatory cytokine mainly secreted by macrophages, binding to its receptors TNFRSF1A/TNFR1 and TNFRSF1B/TNFBR. TNF-α plays a role in regulating immune cells, cell proliferation, differentiation, apoptosis, lipid metabolism, and coagulation. It exists as a multimer of two, three, or five non-covalently linked units. The studies have suggested a neuroprotective function for TNF-α. It is involved in the immune response to bacterial, viral, parasitic, and certain fungal infections, as well as its role in the necrosis of specific tumors. TNF-α induces cytolysis or cytostasis of certain transformed cells, being synergistic with IFN-γ in its cytotoxicity. It has been associated with autoimmune diseases, insulin resistance, and cancer.

In the assay, samples, standards or controls are added to the microplate coated with the primary antibody, and then bind to the immobilized antibody in the microplate wells. Subsequently, the second antibody is introduced to the mixtures as the detector, followed by the addition of a substrate solution that generates signal to be measured. The intensity of this signal correlates to the concentration of human TNF-α in the original specimen.
